P-224: The Effect of Alpha Lipoic Acid on ROS Production and TAC Levels of Cultured Isolated Pre-antral Follicles Drived from Mice Vitrified Ovaries

Abstract:

Background: Ovarian cryopreservation is an efficient way to preserve fertility of cancer patients before chemotherapy. This study was designed to evaluate the effect of apha lipoic acid (ALA) on reactive oxygen species (ROS) production and total antioxidant capacity (TAC) of cultured isolated pre-antral follicles derived from vitrified and non-vitrified ovarian tissue. Materials and Methods: Immature mouse ovaries were vitrified by cryotop method. Pre-antral follicles were isolated and cultured in α- minimal essential medium supplemented with 100 um of ALA. Follicular growth, oocyte maturation were evaluated. Separately, ROS and TAC were measured after 0, 24, 48, 72 and 96 hours of culture with spectrofluorometery and ferric reducing/antioxidant power (FRAP) assay, respectively. Results: The rates of survival, growth, antrom formation and MII oocytes were significantly lower in vitrified groups, whereas in the presence of ALA were significantly higher in both vitrified and non-vitrified groups (p<0.05). ROS production of pre-antral follicles in vitrified and non-vitrified groups was increased, and TAC levels were decreased, while in the presence of ALA, ROS production was decreased and TAC levels were increased after 96 h of culture. Conclusion: ALA improves the in vitro development of pre- antral follicles isolated from vitrified and non-vitrified ovaries through decreasing ROS concentration and increasing TAC level of during the culture period
